DSC directly measures the enthalpy (ΔH) and temperature (Tm) of thermally induced structural transitions in solution. This information gives valuable insights into factors that stabilize or destabilize proteins, nucleic acids, micellar complexes and many other macromolecular systems, alone or as complexes.
This information can be used to develop purification and formulation strategies, predict shelf lives, and characterize and evaluate protein constructs or other biotherapeutic entities. In addition, ligand affinities to a protein target in small molecule drug discovery programs can be ranked rapidly.
The MicroCal VP-Capillary DSC system has an active cell volume of 130 µl, allowing for thermodynamic measurements of precious samples (sample concentrations of about 0.2 mg/ml are typically needed). The system has a standard 96-well plate format for high capacity and loading ease, and up to six 96-well plates can be stored in thermostatically controlled drawers.
The fully integrated autosampler enables running of up to 50 samples per day, with all filling, injection, and cell cleaning functions fully automated for walk-away operation. The system is setup and controlled by the VPViewer control software, and data analysis is performed with MicroCal-enabled Origin software.
