Great for binding kinetiks of proteins

"MicroCal Peaq ITC is easy to use with a fully automated wash module. The integrated analysis software is very helpful with the inbuilt models to detect the mode of binding. The machine and software is easy to use. The equipment will be useful for the researchers to evaluate the protein-protein, protein-nucleic acid, and small molecule interactions. This will be helpful to determine the binding affinity for the any interaction. The analysis will also suggest the mode of binding and the type of binding if it is hydrophobic or ionic interaction. The equipment is fully automated with ease of use. The titration is well controlled and the experiment can be performed at various temperatures."

"The MicroCal PEAQ-ITC is a highly efficient product in terms of providing excellent results and with high accuracy and repeatability. The product is very handy to study DNA-Protein interaction, PNA interactions, etc. very easy to use and less maintenance, ambient temperature is sufficient. As of my knowledge, the product is best available as of now. Very easy to use, requires less time to start titration around 2 minutes. Extraordinary service support. The best product we can get for the money you spend."

  • Application Area:Kinetic and thermodynamic parameters of a protein drug target

"ITC has become my bread and butter instrument for biochemical characterization because it uses small quantities, provides precise, reproducible results, and is label-free. I run a small lab, so getting all of those things for both kinetics and thermodynamics is gold! The drawback of ALL ITC instruments is, of course, how many runs you must do before you find the magic protocol and concentrations, but I will take that over designing multiple, label-dependent assays."

"The MicroCal PEAQ-ITC is a very versatile equipment. Trough ITC experiment it is possibile to obtain a lot of information regarding the thermodynamics, the mechanism and the stoichiometry of the binding between two molecules in solutions. Additionally, you can exploit it in order to describe quantitatively and qualitatively fundamental phenomena underlining nanotechnologies such as the self-assembly of amphiphilic molecules. The instrument is easy to use also by a beginner user and the maintenance is extremely light. Thanks to the automatization of the cleaning step and the relative less time-consuming, it is possible to carry-out up to 12 experiments during the day."

"ITC is a friendly technique, and this instrument has been upgraded, making it quite robust and easy to use. For biochemists, PhD students as well as PostDocs, it represents an excellent source of info for studying protein-small molecules interactions. If your molecules are water soluble or solubility can be achieved by adding a small amount of organic solvent to your buffer solutions, it is the method of choice to obtain thermodynamic parameters for your lead molecules, affinity constant, enthalpy, stoichiometry of interaction and so on... Last but not least, it helps a lot when you want to know if any compounds do their job by interacting directly with your target or not."

Great instrument, good service.

  • Application Area:Protein metal binding

"I like the instrument design. It is easy to operate, all parts seem very sturdy and there are no extra elements. Malvern did a good job writing the manual for this instrument which is helpful for troubleshooting your experiment and also for maintenance. The software is nice and user-friendly, has a lot of functions that can help you analyze your experiment in the best way. The only issue is that the equations used for calculations are not provided in the software, they are discussed in the manual. As a visual person, I would prefer seeing all calculations but that's just me. After installation, we had a few training sessions (one with the service engineer and another with the application specialist) which were definitely enough to start using the instrument with confidence. As for the maintenance, we recently had our annual check and everything was fine. The engineer came for a day, tested and cleaned our instrument. The only issue was the communication with the service engineer as he was not the best in responding and it took some time to schedule his visit. Many thanks to Stoyan who always answers all questions about experiment design and data analyses. His experience definitely helps in troubleshooting."

Mediocre instrument with very few advantages over the VP-ITC.

"Although I appreciate that the low-volume Peaq-ITC allows for rapid testing (~8 experiments per day on average) the overall use of the instrument is miserable. It will randomly disconnect from the computer every once in a while. Luckily this hasn't happened during an experiment, but is frustrating when you go to start an experiment and you have to shut down the computer to get the instrument to reconnect again. Furthermore, although the automatic cleaning is useful, it is the most frustrating aspect of the instrument. Too many parts and seals necessary. Most ridiculous is that you have to wait 7-9 minutes before the cleaning system will tell you there is an error with the cleaning, and will cancel. So you check the connections and restart, wait 7-9 minutes again for it to fail again. You then get tired of waiting for it to clean and do a ridiculous manual clean (there is no good way to do a manual clean), and there will be an error stating the instrument hasn't been cleaned in XX-uses. At the minimum there should be an override control of the cleaning system so experiments can continue. To top it off, the most frequent problem that causes this cleaning issue is part #: ASY5020090. A little tube with a connector and a little black seal for ~$300, which we have to replace every 3-4 months, as that is approximately how long we have until this little part fails and we can no longer utilize the instrument because it will not clean. The sensitivity is high on the instrument, which is good. However, when systems has low heat, the signal-to-noise ratio is very high and the baseline is terrible. This, in turn, makes you use more sample at a higher concentration, wholly defeating the purpose of this instrument. The error between experiments is about 2-3 times higher than the error between experiments on the VP-ITC as well, which is ridiculous. I need to do ~6-8 experiments to obtain the same error on 3 experiments by the VP-ITC. So basically you can do quicker experiments, but you need the same overall amount of time to get reasonable data. Lastly, the data analysis software is of poor quality. The switch to proprietary data analysis has made data analysis cumbersome, compared to the use of Origin as the fitting software. Two-Site fitting is backwards when binding site #1 is actually binding site #2. The fitting curve gets stuck when trying to fit two-site data, and requires extensive manual fitting to get it to work, with questionable results and thermodynamics. Although this is a small complaint, when making the isotherms publication quality, you have to use individual drop-downs for every little piece of the graph. The lines are not even black as the default, but a weird blue-grey color. Changing the raw data to black then the axis labels to black, then switching and changing the fitted data colors takes a ridiculous amount of time. Plus there appears to be ~100 color choices, which is just overkill."

"The MicroCal PEAQ-ITC is a significant upgrade from the VP-ITC and ITC200 in terms of the ease of maintenance. All washing steps are now automated and easy to perform. We find this helps to keep the instrument in an excellent condition even with multiple users. Due to the somewhat complicated cleaning procedures of previous versions of the ITC we found that routine maintenance was not always performed which greatly affected results in this highly sensitive instrument. The PEAQ is still a low volume isothermal titration calorimeter ideal for label-free, in-solution studies of biomolecular interactions. Software upgrades have also made the PEAQ easy to use even for the most inexperienced scientists in the lab, including help in experimental planning and optimisation of parameters prior to performing any runs. We find the PEAQ really useful for confirming binding, measuring stoichiometry, protein QC, troubleshooting other biophysical methods and identifying different mechanisms of binding in closely related compounds."
