Nuclear receptors (NRs) are a superfamily of ligand-dependent transcription factors that control diverse aspects of growth, development and homeostasis, making them exciting and important targets for drug discovery.
The HitHunter™ Nuclear Hormone Receptor Assays are homogeneous immunoassays where analogs to the respective nuclear hormone compete with ED-nuclear hormone for binding to the respective nuclear hormone receptor. The assays were designed using EFC detection with a choice of either chemiluminescent or red-shifted fluorescent intensity detection.
Features and Benefits
» Scaleable
• Protocols for 96-. 384- & 1536-well microplate applications
» Flexible
• EFC detection with a choice of Chemiluminescent or red shifted fluorescent intensity readouts
» Robust
• Large signal to background ratios for superior signal differentiation and assay performance
Measuring Nuclear Hormones with HitHunter EFC Detection
The HitHunter™ Nuclear Hormone Receptor Assay Kit utilizes an ED-steroid hormone conjugate in which the steroid hormone is recognized by the steroid hormone receptor. In the absence of free hormone, the ED fragment is incapable of reassociation with EA to form active enzyme and no signal is generated. In the presence of free Ligand, ED-steroid hormone conjugate competes with the free Ligand for receptor binding. The amount of signal is proportional to the amount of nuclear hormone receptor bound.
