HitHunter™ cGMP Assay by DiscoveRx Corporation

Overview and Description Guanosine 3’5’-cyclic monophosphate (cGMP) is an intracellular second messenger. Either receptor bound natriuretic peptides or nitrous oxide activate the production of cGMP from GTP. Cyclic GMP signaling is mediated by three effector proteins – protein kinase G (PKG), phosphodiesterase 5 (PDE type 5), and cGMP regulated ion channels. Cyclic GMP affects processes in the brain, cardiovascular organs and sensory systems. Features and Benefits » Directly measure cGMP from ce... Read more

Overview and Description
Guanosine 3’5’-cyclic monophosphate (cGMP) is an intracellular second messenger.

Features and Benefits
» Directly measure cGMP from cells in suspension
» Robust
• Large signal to background ratios for superior signal differentiation and assay performance
» Scaleable
• Protocols for 96-. 384- & 1536-well applications
» Reduced fluorescence interference
• Luminescent detection reduces interference from fluorescent compounds, lowering false positives

Measuring cellular cGMP with EFC chemiluminescent detection
HitHunter cGMP is an in-vitro based competitive immunoassay. Free cGMP from cell lysates compete for antibody binding against labeled ED-cGMP conjugate, a small peptide fragment of ß-galactosidase (ß-gal). Unbound ED-cGMP is free to complement with EA, an inactive ß-gal EFC enzyme, to form active enzyme, which subsequently hydrolyzes luminescent substrate. The amount of luminescent signal produced is directly proportional to the amount of free cGMP in the cell lysate.
