"Glioblastoma spheroids of ~1mm in diameter are incubated with Draq7 for 16-20hrs before imaging. Draq7 is visualized in far red and stains dead nuclei, without discriminating cell death pattern. Easily combined with blue and green probes. Photobleaches after 6hrs."

"We use this product to look at nuclei in both 2D and 3D fixed immunofluorescence imaging. Very useful if you do not have access to a 405 laser and cannot use DAPI, as this uses the 647 channel."

"We used DRAQ7 to monitor the cell death along implants over time. For this purpose we required a non-toxic and stable fluorescent dye to detect dead cells. DRAQ7 met all necessary requirements and worked also in the hydrogel. It was very easy to use with immediate results without optimization period. Some other advantages are that it works without UV light and is non-toxic to cells over time."

"My colleagues and I could see a clear and bright signal even with low laser power. There was never an issue of poor dye penetration into 3D cellular aggregates, as would otherwise be observed with more common nuclear dyes, such as Hoechst . The quality of staining is usually consistent across experiments and among different colleagues. This follows that our results were also consistent across experiments. The dye was stable after repeated uses, and the signal intensity of DRAQ7 did not decrease with repeated use. Moreover, as it is simply added to cell culture media, it fits easily into any experimental design. I would recommend DRAQ7 to other researchers for its stability, fluorescence quality and its convenience of use. One suggestion for improvement would be to create versions in other fluorescence colors instead of just one which is presently APC."

"We used DRAQ7 to visualize the formation of a necrotic core in fresh isolated Islets of Langerhans, which are prone to hypoxia. DRAQ7 was simply added to the medium to a final conc. of 1 microM. This compound appeared specific for staining the nuclei of necrotic cells only. It is easy in use, provides a fairly low background and stable regarding bleaching."

Very bright photostable far red dye

"DRAQ7 was used to measure cell death in complement-dependent-cytotoxicity experiment. Our project was to determine the ability of anti-Tn antibody to mediate death of Tn expressing in association with complement. Briefly, Tn expressing cells were incubated with anti-Tn antibody, in the presence of complement and DRAQ7 as a viability marker. Cell were incubated in Incucyte System (www.essenbioscience.com/en/products/incucyte?gclid=CObfsKOZjM8CFWMW0wodVw0JSQ) and the appearance of red fluorescence (staining of dead cells by DRAQ7) was monitored every 30 minutes. We were able to measure cell death over time by the use of DRAQ7. Fluorescence emission signal was high, and the results obtained were reproducible. Among all viability dye we have tested, it was the only one that emits strong signal after several hours of incubation. "

Although the product is commonly used as a FACS marker for cell death, I have found great utility as a way to real time mark cell death using Incucyte. "This has enabled us to do experiments that were not otherwise possible before. Not only is it a much cleaner signal than other viability dyes, but the fact that it is non-toxic has enabled us to do time courses with the same samples. We have performed 3D spheroid experiments over a 4week period, reading plates every 72hours with zero toxicity! It therefore saves us large amounts of money that would have been spent on ending experiments at each time point using toxic dyes. Highly recommended! "

  • Application Area:Live cell screening

"Draq7 is a great dye for labelling cells and is very easy to use. Using this dye we have been able to do live cell screening of cells, as it is non-toxic to cells. However, overall the cost is a bit expensive for screening."

DRAQ7 fluoresces in the far-red – with the same spectral properties that have made DRAQ5™ a reagent of choice in HCS.
DRAQ7™ only labels membrane-compromised dead, dying and apoptotic cells and gives clear discrimination between these and healthy cells in high content cytotoxicity and viability assays and in flow cytometry.

The far-red emission, longer wavelength than TOTO-3, permits better combination with other fluorescent probes. It is spectrally compatible with the commonly used orange/red mitochondrial health probes, and UV-excited vital dyes.

DRAQ7 can be used in RNAi knock-down and long-term, dynamic viability assays to report in vitro cytotoxicity and apoptosis.

Also, DRAQ7 efficiently counterstains fixed cell and tissue preparations. It offers the same differential nucl:cyto staining for dual compartment segmentation in translocation tracking and cell morphometrics for in vitro toxicology, that has made DRAQ5™ a favourite of HCS assay developers.
 
DRAQ7 is supplied as a convenient aqueous, ready-to-use solution. The 1 ml pack size (GBP 140.00 / ca. USD 220.00) is sufficient for at least 200 flow cytometry tests, 300-600 slides and up to 10,000 1536-MTP wells.
