CYP4F3A is a non-hepatic P450 enzyme. Its expression is localized to the ER of polymorphonuclear leukocytes (PMNL). CYP4F3A is a major enzyme involved in the ω-oxidation of Leukotriene B₄ (LTB₄) in humans, yielding the biologically less active 20-hydroxylase product. LTB₄ is a potent chemical activator of inflammatory reactions; the turnover of LTB₄ has been used to determine the extent and duration of inflammation. The substrate specificity of CYP4F3A is shown to include several ecosinoids in addition to LTB₄ such as lipoxin A4 and B5, 12-HETE, and 5-HETE. CYP4F3A and CYP4F3B originate from the same pre-curser mRNA. The two isoforms differ by a single exon (exons 3 and 4), originating from a tissue specific alternative splice mechanism.
