The pGFP2-DEVD-Rluc(h) vector is an apoptosis biosensor that monitors caspase-3 activity in live cells. This vector contains a fusion GFP2-DEVD-Rluc(h) gene which harbors a caspase-3 recognition/cleavage site Asp-Glu-Val-Asp (DEVD in single letter amino acid code) between a modified Green Fluorescent Protein (GFP2) and codon humanized Renilla luciferase (Rluc(h)) coding sequences. The GFP2-DEVD-Rluc(h) fusion protein produces a strong BRET2 signal in the presence of the coelenterazine substrate, DeepBlueC. Once recognized by caspase-3, the expressed fusion protein is cleaved within the DEVD sequence, disrupting the BRET2 signal between Rluc and GFP2. The fusion gene is placed under the control of the cytomegalovirus (CMV) promotor thus assuring a very high constitutive expression in a variety of cells.
