The pGFP2-MCS-Rluc(h) vector allows for the expression of a BRET2 fusion construct that can be used as a biological sensor. The multiple cloning site (MCS) located between the GFP2 and Rluc(h) genes permits the insertion of modulator sequences such as e.g. a protease consensus sequence. This construct can also be used as a positive control though producing a lower BRET2 signal value than the straight fusion between GFP2 and Rluc (part # 6310030). Both GFP2 and Rluc genes have been codon humanized to ensure higher expression levels in mammalian cells. The fusion protein is placed under the control of the cytomegalovirus (CMV) promoter thus assuring a very high constitutive expression level in a variety of cells. |
