ADP Quest™ and ADP Quest HS assays are unique tools for researchers for profiling enzyme activity of kinases and ATPases. The assay employs a generic, non-antibody based, non-radioactive method to detect the amount of ADP produced as a result of enzyme activity. The amount of ADP produced is directly proportional to enzyme phosphotransferase activity and is measured as a robust red-shifted fluorescence emission signal. ADP Quest™ assays are useful tool for determining Michaelis-Menten kinetics and profiling inhibitor mode of action and potencies. The assay is for research use only and has been validated in 96-well and 384-well microplate formats.
ADP Hunter™ Plus and ADP Hunter HS are ideally suited for HTS. The assays tolerate a broad range of ATP concentrations, and use unmodified peptides or whole proteins as substrates. ADP Hunter HS detects as little as 300nM ADP, while ADP Hunter Plus can tolerate up to 200uM ATP. ADP Hunter assays have been designed to minimize compound library interference, and have been developed with a stop solution to stabilize the reaction at a given endpoint for up to 2 hours.
