"Waters has introduced Acquity BEH amide (HILIC Lite) columns parallel to other HILIC columns. BEH Amide columns are far superior in stability and robustness then other HILIC columns in waters portfolio. Generally HILIC columns tend to leech polymer into ion source of Mass spectrometers leading to serious contamination. Also HILIC columns are very sensitive to changes in mobile phase buffers. Both of these problems are addressed quiet well by introduction of this new column. Although still lot more to be done to make such column comparable to a good RP-18 column, still they are probably better choice for studying acid and phosphates such as metabolites of Carbon Central Metabolism. "

"The column very well-kept the reproducibility and the separation robustness even after ten thousand injections. Peak shapes are well-maintained at concentrations very close to lowest limit of quantification, no or very minimum peak smoothing is needed. After-sales cares are very professional and eager to help solving problems. When the column is conjugated with guard column and in-line filter, the life-time and separation reproducibility are ensured. Little expensive in the beginning, but definitely worth it in the long run. "

"BEH columns are useable for all pH range reversed-phase separation and helped us to replace other laborious desalting cleaning steps before LC-MS/MS experiments for peptides. We use it for iTRAQ, phosphopeptide analysis and global proteomic analysis. Peaks are evenly distributed across the gradient unlike strong cation exchange methods. Basic RPLC using BEH column is our favorite offline method. Peptide separation is very different in low and high pH condition offering a scope for better fractionation. The only one pre-step needed is 30 kDa buffer exchange of protein lysate in urea."
