Analysis of Drug Substances in Headache Medicines with the PerkinElmer Flexar FX-15 System Equipped with a PDA Detector
03 Feb 2012: This application note presents a modified USP method for the analysis of aspirin, acetaminophen and caffeine using a superficially porous particle column and a PerkinElmer® UHPLC platform. The changes to the USP method increase throughput and reduce solvent consumption. Method conditions and performance are presented. Two headache medicines, one brand name and one generic are tested and the amount of the active ingredients determined.

Predictive Assays for High Throughput Assessment of Cardiac Toxicity and Drug Safety
03 Feb 2012: In this poster a method is demonstrated for measuring the impact of pharmacological compounds on beating stem-cell derived human cardiomyocytes using the ImageXpress® Micro and the FLIPR® Tetra systems from Molecular Devices, LLC. Both methods allow monitoring of drug impact on the beat rate and amplitude in 96 or 384 well formats. The method demonstrated use of both systems for two important applications: screening compounds for cardiac toxicity, and pre-clinical testing of

Metabolic Pathway Determination Using the XF24 Analyzer
03 Feb 2012: The XF24 Analyzer non-invasively measures physiological changes in cellular bioenergetics and metabolic pathways in vitro using optical biosensors in a 24-well format. Shifts between mitochondrial respiration and glycolysis as a result of genetic, pharmacologic or environmental manipulation can be measured in a single assay the method of which is described in this application note. XF24 assays provide increased throughput in a drug discovery format.

Monitoring Cure Characteristics of a Thermoset Epoxy by ATR/FTIR
03 Feb 2012: A high temperature epoxy curing study is presented as an example application where a high performance, robust ATR accessory is desirable. Through FTIR data, reaction kinetics was determined for demanding and high temperature applications.

Research Automation Solutions by Beckman Coulter Featured at SLAS 2012
03 Feb 2012: Beckman Coulter Life Sciences will offer an array of innovative platforms for cellular and molecular biology research to attendees at this year’s inaugural Society for Laboratory Automation and Screening (SLAS) conference and exhibition at the San Diego Convention Center. An extensive exhibit in booth 619 and detailed workshops will feature automated solutions for high-throughput applications in flow cytometry and the preparation of genomic samples for sequencing.

Measuring Glycolysis and Oxidative Metabolism in Cancer Cells
03 Feb 2012: Learn more about a method for validated OCR and ECAR as indicators of cellular respiration and glycolysis respectively in this application note. By measuring their responses to three well-defined modulators of mitochondrial respiration and glycolysis in the A549 human cancer cell line, it was reported that 2-4 DNP uncouples respiration from ATP synthesis, stimulating both respiration and glycolysis. 2-DG inhibits hexokinase, the first enzyme required for glycolysis, specifically inhibi

Hamilton MICROLAB® NIMBUS Enclosed Workstation Provides Clean and Secure Automation Environment
02 Feb 2012: Hamilton Robotics is pleased to introduce 'MICROLAB NIMBUS Enclosed' at SLAS 2012 – a protected, multifunctional benchtop workstation that enables gripper movements and a range of pipetting capabilities for tubes and plate within a secure environment. The NIMBUS Enclosed accommodates either the Hamilton CO-RE (compressed O-ring expansion) 96-probe multichannel head or four independent 1 mL channels, all within a locked cover. The new enclosure design includes rear expansion and side integration

Screening for Mitochondrial Biogenesis and Toxicity
01 Feb 2012: In this application note, adapted primary cultures of RPTC to exhibit in vivo levels of aerobic metabolism, to be non-glycolytic, and to retain high levels of differentiated functions were obtained. The XF96 Analyzer was used to measure mitochondrial respiration of the RPTC in real time. Changes in carbonylcyanide-4- (trifluoromethoxy) -phenylhydrazone (FCCP)-uncoupled respiration, a measure of electron transport chain (ETC) integrity and a sensitive measure of mitochondrial functional capa

Design and Performance Features of ATR Crystal Plates for Single Reflection ATR Accessories for FTIR
01 Feb 2012: The purpose of this note is to provide comparative information and data to assist in the selection of the best crystal material for specific ATR sampling applications. Factors considered include: throughput, spectral range, crystal material hardness and pH tolerance, and refractive index. Crystal design factors are also considered such as size and how well it accommodates liquids, solids and other sample forms.

Bioenergetic Analysis of Primary Neuronal Deficiencies
01 Feb 2012: Read this application note to learn how the XF Analyzer is designed to handle small sample sizes to monitor mitochondrial respiratory parameters of synaptosomes using 50-fold less protein than previously possible. The results demonstrated a decrease in mitochondrial spare respiratory capacity in bioenergetically compromised synaptosomes that may model deficits characteristic of PD and HD.
