Establishing a Cell Culture Assay Based on Time-Resolved Fluorescence Resonance Energy Transfer (TR-FRET) for Screening G-Protein Coupled Receptors
09 Feb 2010: Time-resolved fluorescence resonance energy transfer (TR-FRET) has become a popular technique in the field of high-throughput screening. This application note demonstrates the TR-FRET kit from Cisbio, the IP-One HTRF assay, under standard high-throughput screening conditions. The IP-One HTRF assay was shown to be a stable cell culture time-resolved fluorescence assay for screening G-Protein coupled receptors.

Functional Screening of Recombinant CHO-M1 Cells using the PHERAstar from BMGLABTECH
09 Feb 2010: This application note demonstrates the HTRF® assay from Cisbio as a fast and reliable method to directly screen a large number of stable transfected single cell clones. Carbachol dose response curves with fresh cells from a growing culture and frozen instant cells measured on BMG LABTECH´s PHERAstar showed similar results for EC50 and signal-to-blank values.

Tag-lite®: New HTRF® Cellular Platform for Cell Surface Receptors’ Study and Screening
09 Feb 2010: In a recent publication, Maurel et al presented the combination of Cisbio’s HTRF and SNAP-and CLIP- Tag technologies to investigate cell-surface protein-protein interaction. In this study, an application for GPCR oligomerization was accurately documented and the advantages of using the Cisbio HTRF-based cell-surface receptor platform presented. Applications such as highly selective and non-radioactive ligand binding assays are described and the 7TM dimerization model can properly be inv

A High Throughput, Label-Free Assay for SDF-1/CXCR4 Signaling
08 Feb 2010: This technical note demonstrates a robust and label-free SDF-1a/CXCR4 BIND assay performed using stem cells endogenously expressing CXCR4. BIND technology allows for cell-based detection of all GPCR subtype activation using a single assay and is sensitive enough for use with both over-expressed and endogenous GPCRs.
